Life Science Buffers
Balanced salt solutions and powders used for molecular, protein, nucleic acid, and cell biology applications including cell culture, electrophoresis, ELISA, and chromatography processes; available in a variety of formulations and grades.
Buffer solutions serve to keep pH (acidity or alkalinity) nearly constant in a variety of chemical and biological applications. For example, a bicarbonate buffer helps maintain the pH of blood, a buffered saline maintains cellular contents at a consistent pH level, and buffers help maintain a narrow pH range for enzymes to function correctly.
Most biological samples used in research are kept at a pH of about 7.4. Common biological buffers used for tissue culture include Dulbeccos phosphate buffered saline (PBS), Tris base, HEPES, MOPS, PIPES, and other formulations developed for specific cell lines or applications. Most buffers used in cell culture are also DNase-, RNase-, and protease-free.
The choice of buffer for a particular biological reaction or site depends on several factors:
- Temperature
- Desired or target pH
- Buffer toxicity (to the system)
- Buffer interactions with other system components

The Tris-EDTA (TE) Buffer is a commonly used buffer solution in molecular biology, especially in processes involving nucleic acids. This buffer is composed of a 20X concentration of Tris, a buffering agent, and EDTA, which has the ability to prevent degradation of DNA and RNA by chelating magnesium or other divalent metal ions, and allows pH regulation within the range of 7.5-8.5. It is widely used often for DNA and RNA gel electrophoresis, and also for DNA automated sequencing.

MES Buffer [2- (N-morpholino) ethanesulfonic acid] is a zwitterionic Good's buffer used in biological and biochemical research. It maintains a stable pH within the range of 5.5 to 7.0 and is highly soluble in water. MES is commonly used as a running buffer for bis-tris gels, chromatography, and in protein purification. It is valuable for enzymatic reactions, and acts as a non-coordinating buffer in metal ion chemistry. It has minimal UV absorbance and is stable in various applications.

MES is one of the so-called Good buffers developed for biological applications. This product is BioPerformance certified (suitable for electrophoresis, tissue culture, and molecular biology applications. A buffer using MES free acid can be prepared by titrating the free acid with sodium hydroxide to the desired pH (pKa/-1). Alternatively, volumes of equimolar solutions of MES free acid and MES sodium can be mixed to attain the desired pH. Standard mixing tables using stock solutions to prepare a buffer of a given pH have been published.
